There are Set's Marks all over the Tombs in Lara Croft and the Temple of Osiris. The evil god Set placed them in the tombs to stop Lara and her companions from reassembling Osiris.
These golden glowing orbs sometimes spawn Enemies like Skeletons or Scarabs or have to be closed to trigger doors. They can only be destroyed by shooting at them with the Staff of Osiris for a longer period of time. Sometimes more than one mark has to be closed at the same time - e.g. by using Mirrors - to progress further in the game. They are also part of the body of the huge snake Lara has to face in the Tomb of the Silversmith.
